DNA via diphenylamine

5% Perchloric acid (w/v)
B
300 mg diphenylamine in 20 ml acetic acid +
0,3 ml conc. sulfuric acid and
0,1 ml 50%iger aqu. acetaldehyde.
(fresh !!)
- The sample is mixed with 1,0 ml of A (up to 1:1)
- The mixture is heated to 70 °C for 15 minutes.
- 1,0 ml with about 5 - 10 µg desoxyribose (30 - 60 µg DNA) is mixed with 2,0 ml of solution B and incubated over night at 30 °C.
- Avoid evaporation!!
- DNA is quantified spectrophotometrically at 600 nm
- Using desoxyribose as standard 1 µg desoxyribose = 6 µg DNA.
