Problem caused by SDS

 

SDS :          -          inhibits digestion by trypsin and endoproteinase Lys-C

                   -          HPLC – separation of peptides

 

e.g. Tryptic digestion of transferrin with traces of SDS

 

Remove SDS via :

 

*       Extensive dialysis

*       Extraction with acetone

        - 450 µl acetone, 1mM HCl, 3h 20 °C

        - centrifugation,

        - wash with 2 x 100 µl acetone

        - dry the pellets with air
