Sequencing of proteins

1.      Characterize the proteins

2.      Purify a sufficient amount 

3.      Determine the amino terminus 

4.      (Determine the carboxy terminus)

5.      Split off acetyl-, formyl-,... - groups

6.      Digest the protein into peptides

7.      Purify the peptides 

8.      Sequence  via sequenator or by hand

9.      Get sequence from overlapping peptides
